摘要
建立了高效液相色谱-蒸发光散射检测器(ELSD)快捷测定、控制黄姜薯蓣皂甙元产品纯度的方法。采用色谱柱:ZORBAX SB-C18(4.6mm×150mm,5μm),流动相V(水)∶V(乙腈)=10∶90(等梯度),柱温28℃,进样量10μL。ELSD:气流流速1.2mL/min,漂移管温度80℃。在0.1~0.4mg/mL范围内,薯蓣皂甙元峰面积与其质量浓度呈良好的线性关系,R2=0.996,5次RSD 1.2%。与常规应用的HPLC分析方法对比表明,利用HPLC-ELSD检测更简单、快速、准确,学生更加能理解HPLC测试方法。
To establish a high performance liquid chromatography-evaporative light scattering detection(HPLC-ELSD)method for the rapid determination of diosgenin and to provide an effective approach for quality control of product from Dioscorea zingiberensis C.H.Wright,a Dionex p680-AllTECH ELSD2000is applied to measure products.The ZORBAX SB-C18column(4.6×150mm,5μm)is used with water-acetonitrile(V(90)∶V(10))as the mobile phase at temperature 28℃and injecting sample size 10μl.AllTECH ELSD2000 is connected to this HPLC equipment with drift tube temperature of 80℃and nitrogen gas flow-rate of 1.2l/ min.The results show that the calibration curve is linear in the range of 0.1~0.4mg/ml(R2=99.6%).RSD is 1.2%.At the same time,a normal HPLC method is applied to measure the product too and the results prove that the HPLC-ELSD method is simple,rapid and accurate.That can make students understand HPLC method deeply.
