摘要
目的探讨甲状旁腺激素(PTH)对人主动脉平滑肌细胞凋亡的作用,及白藜芦醇(Resveratrol,RES)在其中的作用。方法实验一:体外培养人主动脉平滑肌细胞,用不同浓度甲状旁腺激素(阴性对照组、10^(-6)mol/L、10^(-8)mol/L、10^(-10)mol/L)刺激72 h,用流式细胞仪检测各组细胞凋亡情况,寻找可引起细胞凋亡的最适PTH浓度。实验二:用该浓度PTH及不同浓度白藜芦醇(阴性对照组、10μmol/L、50μmol/L、100μmol/L)共刺激人主动脉平滑肌细胞72h,用流式细胞仪检测各组细胞凋亡情况。结果实验一:相较于对照组,不同浓度PTH刺激下,各组均产生了明显的细胞凋亡,差异具有统计学意义(P <0. 05)。确定10^(-10)mol/L PTH为可诱导细胞凋亡的较低浓度PTH。实验二:用10^(-10)mol/L PTH及不同浓度白藜芦醇(阴性对照组、10μmol/L、50μmol/L、100μmol/L)共刺激人主动脉平滑肌细胞72h,发现相较于对照组,各浓度白藜芦醇(10、50、100μmol/L)均可以减少细胞凋亡,差异具有统计学意义(P <0. 05)。结论一定浓度的甲状旁腺激素可以诱导人主动脉平滑肌细胞凋亡,白藜芦醇对其有抑制作用。
Objective To investigate the effect of resveratrol on parathyroid hormone(PTH)induced apoptosis of human aortic smooth muscle cells.Methods Human aortic smooth muscle cells were cultured in vitro and stimulated with different concentrations of PTH[0(negative control group),10-6 mol/L,10-8 mol/L,10-10 mol/L]for 72 h.Apoptosis of each group was detected by flow cytometry.Optimal PTH concentration that could induce apoptosis was found.Human aortic smooth muscle cells were co-stimulated with PTH and resveratrol[0(negative control group),10μmol/L,50μmol/L,100μmol/L]for 72 h.Apoptosis was detected by flow cytometry.Results Compared with control group,different concentrations of PTH resulted in different apoptosis rates(P<0.05).The lower concentration that could induce apoptosis was 10-10 mol/L PTH and was used for further co-stimulation.Compared with control group,different concentrations of resveratrol reduced apoptosis rate variously(P<0.05).Conclusion Parathyroid hormone can induce apoptosis of human aortic smooth muscle cells.Resveratrol can inhibit the process.
