CXCR4趋化因子拮抗剂AMD3100对人羊水干细胞抑制作用的体外实验研究 被引量:3

摘要 目的通过体外实验研究CXCR4趋化因子拮抗剂AMD3100对人羊水干细胞(h AFSC)的影响。方法羊水干细胞分为对照组(PBS处理)和实验组(AMD3100处理10μg/L)。采用Transwell小室分析细胞迁移力,MTT法检测AMD3100对hAFSC存活的影响,明胶贴壁法测定AMD3100对hAFSC细胞粘附能力影响。结果对照组和实验组hAFSC迁移细胞数分别为80. 35±3. 70和36. 50±3. 66,粘附细胞数分别为31. 48±3. 21和11. 41±1. 89,存活细胞OD值分别为0. 91±0. 04和0. 36±0. 02,实验组均显著低于对照组,差异均有显著性(P <0. 05)。结论 AMD3100在体外实验中抑制羊水干细胞的迁移、粘附和存活,其作用机制可能涉及SDF-1/CXCR4。 Objective Inhibition of human amniotic fluid stem cells(hAFSC)by AMD3100 was studied in vitro.Methods Amniotic fluid stem cells were divided into control group(by PBS)and experimental group(by AMD3100 10μg/L).Cell migration was analyzed using a Transwell chamber.The effect of AMD3100 on the survival of hAFSC was examined by MTT assay.The effect of AMD3100 on the adhesion of hAFSC cells was determined by gelatin adherence method.Results In the control group and the experimental group,the number of hAFSC migration cells were 80.35±3.70 and 36.50±3.66,respectively,and the number of adherent cells were 31.48±3.21 and 11.41±1.89,respectively.The OD values of viable cells were 0.91±0.04 and 0.36±0.02,respectively.Group experiments were significantly different(P<0.05).Conclusion AMD3100 inhibits the migration,adhesion,and survival of amniotic fluid stem cells in vitro,and its mechanism may involve SDF-1/CXCR4.
