三聚氰胺及其与三聚氰酸联合攻毒对Vero细胞的毒害作用研究 被引量:1

摘要 为补充三聚氰胺的毒理学资料,以易于生长的传代细胞为研究对象,采用形态学观察、MTT法、中性红法等,检测三聚氰胺及其与三聚氰酸混合物对体外培养的Vero细胞的影响。结果表明:不同浓度三聚氰胺及其与三聚氰胺混合物对Vero细胞均会产生一定影响:圆缩细胞增多,贴壁细胞数量减少,且呈现出一定的浓度依赖性。混合组还表现出不规则细胞形态增多。三聚氰胺浓度超过0.75mg·mL-1时会极显著抑制细胞的增殖(p<0.01),细胞毒性级为1级;而二者混合攻毒时毒性明显增强,浓度超过0.094mg·mL-1时会显著或极显著抑制细胞的增殖(p<0.05或p<0.01),细胞毒性级为2级。同时,该类化合物对蛋白质的合成没有明显的影响,但三聚氰胺及其混合物,均会干扰Vero细胞对中性红的摄取率,由此推测该类化合物对细胞的溶酶体可能会产生一定的损伤。三聚氰胺及其混合物对Vero细胞均会产生一定毒性。 Melamine and cyanuric acid have attracted worldwide attention due to their potential harm. Therefore, we carried out this study, aiming to supplement the existing toxic profile of melamine and its homologues, and to provide scientific data in order to establish the monitoring system of food safety to such compounds. In the present study, with easy growing passaged cells as the material, we investigated the effect of melamine in the presence and absence of cyanuric acid on cultured Vero cells in vitro by several techniques including morphological observation, MTT, and neutral red methods. The effects of different concentration melamine in the presence and absence of cyanuric acid on Vero cells were observed. An increasing in round shrinking cells,and a decrease in adherent cells showing a concentration-dependent pattern. Moreover, combined group of melamine and cyanuric acid showed an increase in irregular morphology cells. Melamine could inhibit significantly the cell proliferation when its concentration over 0.75 mg/ml(p<0.01), exhibiting the 1 grade of cytotoxicity. Also, an increasing cytotoxicity was observed in combined group of melamine and cyanuric acid. Combination of melamine and cyanuric acid could inhibit significantly the cell proliferation when its concentration over 0.094 mg/ml(p<0.01 or p<0.05), exhibiting the 2 grade of cytotoxicity. No obvious effect was observed in protein synthesis, however, melamine in the presence and absence of cyanuric acid could interference uptake rate of Vero cell to neutral red. Therefore, we speculated that these compounds could lead to certain damage to cell lysosomes.These results obtained from this study suggested that melamine in the presence and absence of cyanuric acid has certain toxicity on Vero cells.
