表面等离子共振传感器检测转基因植物中苏云金芽孢杆菌cry1Ac蛋白的方法研究 被引量:1

摘要 为建立表面等离子共振传感器检测转基因植物中苏云金芽孢杆菌cry1Ac蛋白的方法,根据生物分子间的相互作用,在金片表面修饰cry1Ac蛋白的特异性单克隆抗体,对不同稀释梯度的cry1Ac蛋白进行检测研究。结果表明:该方法可有效的检测到cry1Ac蛋白,检测灵敏度可达到10ng·mL-1,与cry2A蛋白及CP4-EPSPS蛋白无交叉反应。说明利用表面等离子共振方法检测cry1Ac蛋白操作简单,省时且灵敏度高,特异性强,可用于对cry1Ac蛋白的定量检测。 The objective of the study was to establish a detection method of Bacillus thuringiensis(Bt) cry1 Ac protein by surface plasmon resonance(SPR) sensor and provide a new way for the cry1 Ac protein detection. Using SPR technology and biological molecules interaction analysis principle, the specific monoclonal antibody on the gold surface for the research of cry1 Ac protein detection was modified. The limit of detection of cry1 Ac protein could be 10ng·mL-1by this method and it had a good specificity. The method of SPR detection of cry1 Ac protein was very simple, rapid and high sensitivity. It could be used for the qualitative detection of cry1 Ac protein and provide a new detecting method for cry1 Ac protein detection. It could has a broad application prospect in Bt gene-modified plant detection.
