摘要
苯硫脲尝味实验是遗传学实验经典内容,实验内容主要通过收集口腔黏膜上皮细胞,采用50 mmol/L NaOH和1 mol/L Tris (pH8. 0)直接制备DNA模板,选择合适PCR引物扩增出TAS2R38苦味基因,使用内切酶Hae III酶切扩增产物,从而区分出不同基因型。结果表明,该方法解决了上述问题,简化了实验流程,激发了学生学习兴趣,取得了良好的教学效果。
The phenylthiocarbamide tasting was a classical genetic experiment.The new program is updated with collection of oral mucosa epithelial cells,direct preparation of DNA template uses 50 mol/L NaOH and 1 mol/L Tris(pH 8.0),suitable PCR primers are selected for TAS2 R38 gene,digestion and genotyping are completed by restriction endonuclease Hae III.The results indicate that the new method has solved the problems,simplified the process,stimulated students’interest,and achieved better results.Meanwhile,the experiment requires conventional instruments and reagents,which is convenient for other universities to carry out their experimental teaching work.
