摘要
目的通过检测高脂血症(HLP)大鼠用药前后血浆中硒代甲硫氨酸(SeMet)、硒代半胱氨酸(SeCys)、亚硒酸盐(Se(IV))、硒酸盐(Se(VI))和甲基硒代半胱氨酸(SeMe Cys)不同形态的硒化合物,确定高脂血症与不同形态硒化合物的相关性。方法通过蛋白质沉淀和酶水解方法分别从血清样品中提取分析物。采用HPLC-ICP/MS方法,同时测定大鼠血浆样品中的硒化合物,包括硒代甲硫氨酸(SeMet)、硒代半胱氨酸(SeCys)、四价亚硒酸盐(Se (IV))、六价亚硒酸盐(Se(VI))和甲基硒代半胱氨酸(SeMe Cys)。结果采用沉淀蛋白法处理样品,可以检测到Se(IV),采用酶水解法可以检测到SeCys。高脂血症组与正常对照组相比,大鼠血清中Se(IV)与SeCys含量有明显降低,高脂血症组大鼠用西药阿托伐他汀钙片以及中药绞股蓝总苷治疗四周后,血脂水平降低,Se(IV)与SeCys均有不同程度的恢复。结论该方法证明了HLP大鼠血清中硒化合物的变化,Se(IV)与SeCys可以作为临床监测高脂血症的重要的指标化合物。
Objective To detect the levels of SeMet,SeCys,Se( IV) and Se( IV) in plasma of hyperlipidemic rats before and after treatment,selenium( Se( VI)) and selenocysteine( SeMeCys),were used to determine the association between hyperlipidemia and different selenium compounds. Methods The analytes were extracted from serum samples by protein precipitation and enzymatic hydrolysis,respectively. Simultaneous determination of selenium compounds in rat plasma samples including SeMet,SeCys and Se( IV) selenium by HPLC-ICP/MS,hexavalent selenite( Se( VI)) and methylselenocysteine( SeMeCys). Results In the four plasma samples,Se( IV) was detected by the sedimentation protein method,and SeCys was detected by enzymatic hydrolysis. Compared with the normal control group,the content of Se( IV) and SeCys in the plasma of rats in the hyperlipidemia group was significantly decreased,and Se( IV) and SeCys in the hyperlipidemia group were recovered to some extent. Conclusion This method proves the changes of selenium compounds in serum of HLP rats. Se( IV) and SeCys can be used as important index compounds to prevent hyperlipidemia.
