RP-HPLC-UV法同时测定毛大丁草中大丁苷和8-甲氧基补骨脂素的含量 被引量:5

摘要 目的建立HPLC方法同时测定毛大丁草(Gerbera piloselloides(L.) Cass.)中大丁苷和8-甲氧基补骨脂素的含量。方法采用WondaCract ODS-2色谱柱(250 mm×4.6 mm,5μm),使用乙腈(A)-水(B)溶液为流动相,进行梯度洗脱,流速:1.0 mL·min^(-1),检测波长:282 nm,柱温:25℃。结果大丁苷和8-甲氧基补骨脂素的标准曲线线性关系良好,相关系数r分别为0.999 3和0.999 4。实验的精密度、重复性和稳定性均能达到要求,大丁苷的平均回收率为99.30%、8-甲氧基补骨脂素的平均回收率为99.40%。以上2种成分都可以与基线分离,并且无其他成分峰的干扰。结论本方法适用于毛大丁草药材的质量控制。 Objective To develop a method of high performance liquid chromatography for the content determination of Gerberinside and 8-Methoxypsoralen in Gerbera piloselloides(L.) Cass..Methods The analysis was performed on a WondaCract ODS-2 column(250 mm×4.6 mm,5 μm) with gradient elution.The mobile phase was water(B) and acetonitrile(A).The detection wavelength was 282 nm.The column temperature was set at 25 ℃.Results Gerberinside and 8-methoxypsoralen were separated well with a good linearity,the r values were 0.999 3 and 0.999 4,respectively.The precision,stability and repeatability of the experiment all meet the requirements.The average recovery rate of gerberinside was 99.30%,the average recovery rate of 8-methoxypsoralen was 99.40%.The above two composition could be separated from the baseline,and they had no interference from other component peaks.Conclusion The method has the advantages of simple steps,fast analytical speed and strong specificity,and can be used for the quality control of Gerbera piloselloides(L.) Cass..
