一种纳米二氧化硅组织粘合剂的制备与初步评价

摘要 目的设计并制备具有较高比表面积的二氧化硅纳米粒,初步探究其对生物组织的粘合效果。方法以正辛烷作为油相,以十六烷基三甲基溴化铵为表面活性剂,构建一种水包油的非均相反应体系;在该体系中,以正硅酸乙酯为硅源,以L-赖氨酸为水解缩合催化剂,制备二氧化硅纳米粒;采用扫描电镜、动态激光散射仪、比表面积和孔径分析仪对所合成的纳米粒进行表征。以N,N-二甲基丙烯酰胺为单体,过硫酸钾、四甲基乙二胺作为氧化还原引发剂,N,N-亚甲基双丙烯酰胺作为交联剂,制备聚N,N-二甲基丙烯酰胺仿生水凝胶;使用市售、自制二氧化硅纳米粒粘合聚N,N-二甲基丙烯酰胺仿生水凝胶以及Wistar大鼠右侧肝叶截面,采用机械牵拉的方法测试不同比表面积纳米粒对聚N,N-二甲基丙烯酰胺仿生水凝胶及大鼠肝脏的粘合能力。结果制备出比表面积为493.1 m^2·g^(-1)的二氧化硅纳米粒,其对聚N,N-二甲基丙烯酰胺水凝胶和大鼠肝脏的粘合力分别为8.63×10^(-3) N·cm^(-2)·mg^(-1)和8.17×10^(-3) N·cm^(-2)·mg^(-1),显著高于比表面积为98.9 m^2·g^(-1)市售二氧化硅纳米粒的粘合能力。结论初步研究结果表明:具有高比表面积二氧化硅纳米粒可用于对生物组织的粘合,比表面积越大,对仿生凝胶和生物组织粘合能力越强。 Objective To design and prepare a kind of silica nanoparticles with large specific surface area and explore its tissue adhesive effect.Methods n-Octane was used as oil phase and CTAB as surfactant to prepare oil-in-water heterogeneous reaction system.Silica nanoparticles were prepared by using ethyl orthosilicate as oil phase and L-lysine as catalyst.Scanning electron microscope,dynamic laser scatterometer,specific surface area and pore size distribution analyzer and other instruments were used to characterize these nanoparticles.DMA was chosen as monomer.KPS and TEMED were used as redox initiator and MBA was used as crosslinking agent to prepare PDMA bionic hydrogel.The mechanical strain was measured to test the adhesion power of the commercially available or self-made silica nanoparticles against PDMA and mouse liver.Results Silica nanoparticles with specific surface area of 493.1 m2·g-1 was prepared.It had better binding ability to PDMA and rat liver than the commercially available silica nanoparticles whose specific surface area was only 98.9 m2·g-1.Conclusion Silica nanoparticles with high specific surface area can be used to bind biological tissues.The larger the specific surface area of the particles,the stronger the binding ability to the biomimetic gels and biological tissues.
