基于UPLC-Q/TOF MS对不同产地东北铁线莲化学成分的比较 被引量:5

摘要 目的明确东北铁线莲药材中是否含有肾毒性成分马兜铃酸Ⅰ,不同产地药材在化学成分上是否存在差异。方法采用UPLC-Q/TOF MS方法,通过与马兜铃酸Ⅰ的保留时间、质谱裂解行为进行比对,确定东北铁线莲药材中是否含有马兜铃酸Ⅰ,通过比对不同产地药材的总离子流色谱图,确定不同产地样品在化学成分上的差异。结果产于辽宁、吉林与朝鲜的东北铁线莲样品中均不含有马兜铃酸Ⅰ成分,同时木兰花碱在吉林和朝鲜样品中含量较高,在辽宁样品中含量却较低。结论 UPLC-Q/TOF MS检测方法能够快速分析东北铁线莲药材中是否含有马兜铃酸Ⅰ,木兰花碱的丰度能够区分辽宁与其他产地的东北铁线莲药材样品。 Objective To identify the nephrotoxic component aristolochic acidⅠin the radix and rhizome of Clematis manshurica and to investigate its regional differences in the chemoprofile.Methods An UPLC-Q/TOF MS method was applied,and aristolochic acidⅠwas identified by comparing retention time and mass spectrometry behavior of samples and standard chemical.What′s more,chemoprofile differences were investigated by comparing the EIC of samples.Results Experiment results showed that Clematis manshurica harvested from Liaoning,Jilin provinces and Korea did not contain aristolochic acidⅠ.Another component,magnoflorine,was found with the high content in samples from Jilin province and Korea,while samples harvested from Liaoning province had a relatively low content.Conclusion Aristolochic acidsⅠfrom the radix and rhizomes of Clematis manshurica could be rapidly identified by the UPLC-Q/TOF MS method reported in this manuscript.The component magnoflorine could be used to identify the original place of Clematis manshurica.
