摘要
目的研究款冬酮抗过敏性鼻炎的作用并探讨其作用机制。方法采用质量分数5%2,4-二硝基氯苯丙酮液制备迟发型超敏反应小鼠模型,通过比较小鼠耳肿胀程度以及脾指数和胸腺指数,评价款冬酮的抗炎作用;采用卵白蛋白(ovalbumin,OVA)制备过敏性鼻炎小鼠模型,经灌胃给药和滴鼻给药,通过比较小鼠鼻部症状以及小鼠体内组胺和NO含量变化,评价款冬酮抗过敏性鼻炎的作用。结果款冬酮高、中、低剂量组对迟发型超敏反应所引起的小鼠耳肿均有一定的抑制作用,抑制率分别为51.78%、48.59%和21.87%,呈现一定的量效相关性,并能显著降低胸腺指数。款冬酮高、中剂量组经灌胃给药能显著抑制OVA诱导的小鼠过敏性鼻炎,减少喷嚏、挠鼻和流涕等过敏症状。血清检测结果显示,款冬酮高、中剂量组有降低组胺含量的作用。款冬酮高、中剂量组经滴鼻给药能显著抑制OVA诱导的小鼠过敏性鼻炎,款冬酮高剂量组对过敏性鼻炎症状的抑制效果与阳性药氯雷他定相近。结论款冬酮具有抗炎、抗过敏性鼻炎作用。
Objective To study the effect of ketonone on allergic rhinitis and its mechanism.Methods Delayed type hypersensitivity mouse model was prepared by 5%2,4-two nitrochlorobenzene acetone solution.The anti-inflammatory effect of TUSSILAGONE was evaluated by comparing the degree of mouse ear swelling,spleen index and thymus index.Mouse model of allergic rhinitis was prepared by ovalbumin(OVA).The effect of Tussilagone on allergic rhinitis was evaluated through the changes of histamine and NO contents in allergic mice which were investigated by intragastric administration and intranasal administration.Results The high,middle and low dose group of Tussilagone had certain inhibitory effects on the mice ear swelling caused by delayed hypersensitivity,and the inhibition rates were 51.78%,48.59%and 21.87%respectively,showing a certain dose effect correlation and they could significantly reduce the thymus index.High and medium dose group of TUSSILAGONE through intragastric administration could significantly inhibit OVA induced allergic rhinitis and reduce sneezing,scratching nose,runny nose and other allergic symptoms.The serum test results showed that high and medium dose group of TUSSILAGONE could decreased the content of histamine.High and medium dose group of TUSSILAGONE through intranasal administration could significantly inhibit OVA induced allergic rhinitis.The inhibitory effect of high dose group of TUSSILAGONE on symptoms of allergic rhinitis were similar to positive drug loratadine.Conclusion Tussilagone has anti allergic effects clear,worthy of further study on chemical synthesis.
