摘要
目的:探讨BRMS1mRNA在乳腺癌中的表达及其与乳腺癌临床病理特征的关系。方法:采用Trizol提取组织总RNA,将mRNA转录成cDNA,然后用实时荧光定量聚合酶链反应(RFQ-PCR)技术检测乳腺组织BRMS1mR-NA表达,以BRMS1mRNA和GAPDHmRNA含量的比值表示BRMS1表达水平。结果:乳腺癌组织中BRMS1mRNA的表达水平明显低于癌旁组织(P<0.01),也明显低于正常乳腺组织(P<0.01),BRMR1mRNA低表达与临床病理分期和淋巴结转移有关(分别为P<0.05,P<0.01),而与患者年龄、癌肿大小、病理类型无关(P>0.05)。结论:乳腺癌组织中BRMS1mRNA的低表达与乳腺癌的发展与转移有关,可能成为判断乳腺癌转移的一个标志物。
Objective To study BRMS1 gene expression in breast cancer and itsclinical significance.Methods Total RNA was extracted with Trizol and mRNA wastranscribed reversely into cDNA,then RFQ-PCR was used to detecte BRMS1mRNA in breasttissue,BRMS1 gene expression were presented as the ratios of 3RMS1mRNA to GAPDHmRNA.Results The difference in BRMS1mRNA expressions cancer and its nearby tissue wassignificant(P<0.01=and the expression was significantly lower at cancer tissuethan at the normal tissue(P<0.01)BRMS1 low expression was related to patho logicgrade and lymph node metastases(P<0.05,P<0.01 respectively),but not to age,tumorsize,pathologic type(P>0.05),Conclusion BRMSI low expression may play a crucialrole in the progression of breast cancer and lymph node metastasis,it may be a markerfor judging breast cancer metastasis.
