摘要
[目的]观察高效液相色谱法测定葛根指纹图谱及葛根素、大豆苷效果。[方法]采用高效液相色谱法,HypersilC18色谱柱(250mm×4.6mm,5.0μm),流动相:甲醇:水进行梯度洗脱,流速1.0mL/min,检测波长:247nm,柱温:25℃,建立葛根指纹图谱,测定葛根素、大豆苷含量。[结果]运用梯度洗脱法得到色谱图各个化学成分色谱峰分离度均较好,达到指纹图谱要求。[结论]HPLC指纹图谱鉴别葛根及有效成分,重复性好、特征性强、检测方法简便,为葛根指纹图谱鉴别及质量控制提供方法学依据。
[Objective]To study the traditional Chinese medicine Pueraria fingerprints and quality control of several chemical drug.[ Methods]They were taken with high performance liquid chromatography,HypersilC18 column(250mm × 4.6mm,5.0μm),mobile phase: methanol: water gradient elution,the flow rate was 1.0mL/min,detection wavelength: 247nm,column temperature: 25 ℃ establishment of traditional Chinese medicine Pueraria fingerprint.The fingerprinting analysis was used to determine the content of puerarin,and daidzein,and provide the basis for quality control.[ Results]The degree of separation of the peaks of the various chemical components of the chromatogram obtained by gradient elution method claim reached fingerprint was better.[ Conclusion]HPLC method can be obtained the Radix fingerprint,and its stability,reliability and repeatability are good,can be more fully reflect the quality of Pueraria herbs for the quality control of Pueraria.
