茜芷胶囊对人离体子宫内膜细胞生长的影响

Influences of Qianzhi Capsule on the Growth of Endometrial Cells In Vitro

目的:探讨茜芷胶囊对离体培养的人子宫内膜基质细胞及腺上皮细胞的生长、细胞因子分泌及相关基因表达的影响。方法:将人子宫内膜组织进行体外分离培养,采用MTT、ELISA及半定量PCR方法测定经茜芷胶囊(0 g/L、1×10-2 g/L、1×10-4 g/L、1×10-6 g/L、1×10-8 g/L)作用后基质细胞及腺上皮细胞生长情况及相关细胞因子、基因的表达情况;采用荧光标记法检测茜芷胶囊作用后腺上皮细胞内Ca2+含量。结果:茜芷胶囊在1×10-6 g/L浓度下作用48 h后对子宫内膜腺上皮细胞的生长具有促进作用,并能诱导腺上皮细胞降钙素(CT)m RNA的表达,显著提高腺上皮细胞内Ca2+含量,但对细胞因子的分泌与未经茜芷胶囊处理的对照组比无统计学差异(P>0.05)。结论:茜芷胶囊能促进离体培养的人子宫内膜腺上皮细胞生长,诱导CT m RNA的表达,并显著增加细胞内Ca2+含量,从而改善子宫内膜容受性,而对子宫内膜相关细胞因子的分泌无显著影响。

Objective: To study the influence of Qianzhi capsule on the cell growth, cytokines secretion and the related gene expression in human endometrial stromal cells and glandular epithelial cells in vitro. Methods: The human endometrial tissue was cultured in vitro. The role of Qianzhi capsule(0 g/L,1 ×10-2 g/L, 1 ×10-4 g/L, 1 × 10-6 g/L, 1 ×10-8 g/L) on cell growth, genes expression and cytokines secretion in stromal cells and glandular epithelial cells was determined by MTT, ELISA and semi-quantitative PCR. The Ca2+ content in endometrial glandular epithelial cells treated with Qianzhi capsule was also detected by the fluorescent labeling. Results: Qianzhi capsule promoted the growth of endometrial glandular epithelial cells in the concentration of 1 × 10-6 g/L after incubation of 48 h. It can induce the m RNA expression of calcitonin(CT) of endometrial glandular epithelial cells and significantly increase Ca2+ content of endometrial glandular epithelial cells. Meanwhile, it had no significant effects on the secretion of cytokines compared with the control(P>0.05). Conclusion: Qianzhi capsule can promote the growth of endometrial glandular epithelial cell in vitro, induce the m RNA expression of CT and significantly increase the Ca2+ content of endometrial glandular epithelial cells. Meanwhile, Qianzhi has no obvious effect on the secretion of cytokines.
