摘要
目的探讨全真一气汤影响COPD大鼠肺组织氧化/抗氧化失衡的机制。方法建立COPD大鼠模型,以全真一气汤干预后,实时荧光定量PCR检测肺组织HMOX-1表达水平的变化,Western blot检测肺组织中HMOX-1蛋白水平的变化。结果中药组较模型组HMOX-1mRNA表达增强,其表达丰度分别为模型组的3.33;中药组较模型组HMOX-1蛋白表达上调。结论提示COPD大鼠肺组织HMOX-1酶表达下降,这可能是COPD大鼠肺组织氧化/抗氧化失衡的重要因素,全真一气汤能通过增强HMOX-1的表达及活化,对COPD大鼠肺组织氧化/抗氧化的失衡起到一定调衡作用。
Objective To investigate the mechanism of Quanzheng Yiqi Decoction on the oxidation/antioxidation imbalance in lung tissue of COPD rats.Methods A rat model of COPD was established.After intervention with Quanzheng Yiqi Decoction,the expression of Homo sapiens Oxygenase-1(HMX-1)in lung tissue was detected by real-time fluorescent quantitative PCR.Western blot was used to detect the changes of Homo sapiens(Hox)-1 protein level in lung tissue.Results The mRNA expression of HMOX-1 in the traditional Chinese medicine group was higher than that in the model group,and its expression abundance was 3.33 in the model group;2.The expression of HMOX-1 protein in the Chinese medicine group was up-regulated compared with the model group.Conclusion It is suggested that the expression of HMOX-1 enzyme in lung tissue of COPD rats is decreased.This may be an important factor in the oxidation/antioxidation imbalance of COPD rat lung.Quanzheng Yiqi Decoction can increase the expression and activation of HMOX-1,which is important for COPD.Oxidation/antioxidation imbalance in rat lung tissue plays a role in balance.
