摘要
目的建立当归饮片HPLC指纹图谱,为评价饮片质量提供依据。方法采用HPLC对来自岷县不同产区的50批次当归标准化自制饮片和15批次市售饮片建立相应的指纹图谱,色谱条件为Waters Symmetry®C_(18)(4.6mm×250 mm,5μm)色谱柱,以乙腈-0.5%醋酸水为流动相进行梯度洗脱,流速1 ml·min^(-1),检测波长为254 nm,柱温30℃。利用相似度评价、聚类分析及主成分分析方法对65批饮片共有峰的峰面积进行分析处理。结果建立了当归饮片HPLC指纹图谱,确定了29个共有峰,对7个共有峰进行了化学成分确认;市售饮片相似度普遍低于自制饮片,50批标准化自制饮片不同地理位置间相似度具有一定的差异;聚类分析可大致将不同产区的当归样品分开,市售饮片与标准化自制饮片能够明显区分;主成分分析结果表明岷县秦许镇产当归质量较佳。结论当归饮片HPLC指纹图谱的建立及化学模式识别的应用为饮片的质量控制提供了简单有效的评价方法。
Objective To establish the HPLC fingerprint of Angelica sinensis(Oliv.)Diels,and provide scientific basis for the evaluation of the quality of decoction pieces.Methods The fingerprints of Angelica sinensis(Oliv.)Diels were established by HPLC.All samples were analyzed by Waters Symmetry?C18(4.6 mm×250 mm,5μm)column maintained at 30°C and eluted with acetonitrile and 0.5%acetic acid at the flow rate of 1 mL·min-1.And the detection wavelength was set at 254 nm.The peak areas of common peaks of 65 batches were processed by similarity evaluation,cluster analysis and principal component analysis.Results The HPLC fingerprints of Angelica sinensis(Oliv.)Diels were obtained and 29 common peaks were designated,and 7 main compounds were identified.The similarity of commercially available decoction pieces was lower than that of homemade decoction pieces.Cluster analysis can roughly separate the samples of Angelica sinensis from different production areas.Commercially available decoction pieces and homemade decoction pieces can also be separated by cluster analysis.Principal component analysis showed a higher level of sample from Qinxu town of Minxian county.Conclusion The establishment of HPLC fingerprint of Angelica sinensis(Oliv.)Diels and the application of chemical pattern recognition provide a simple and effective method for the quality control of Angelica sinensis(Oliv.)Diels.
