刺山柑乙酸乙酯部位对APAP诱导肝损伤的保护作用及体外抗氧化活性测定 被引量:3

The effect of ethyl acetate of Capparis spinosal L. against APAP-induced hepatic injury and its antioxidant activity in vitro

摘要 目的探讨刺山柑乙酸乙酯部位(CSEA)对对乙酰氨基酚(APAP)诱导的肝损伤的保护作用及体外抗氧化能力。方法小鼠20只雄性KM小鼠随机分为对照组,模型组,CSEA低,高剂量组(50和200 mg·kg^(-1))。除对照组外其它小鼠给药7天后,腹腔注射APAP(300 mg/kg)建立肝损伤模型。检测生化指标ALT、AST活性以及氧化应激指标T-SOD、MDA、GSH和T-AOC的含量;酶联免疫法(ELISA)法检测肝组织中TNF-α、IL-6和IL-1β含量;HE染色观察肝脏病理学改变;DPPH自由基和羟自由基清除实验检测CSEA的体外抗氧化活性。结果 CSEA能显著降低ALT、AST、MDA、TNF-α、IL-1β和IL-6水平,增加T-SOD、GSH和T-AOC水平,改善肝组织病理变化;CSEA对DPPH自由基和羟自由基有比较高的清除作用。结论 CSEA有较好的体外抗氧化能力并且对APAP诱导的肝损伤具有保护作用,其机制可能与抗氧化和抗炎作用有关。 Objective To investigate the protective effects of ethyl acetate fraction of Capparis spinosal L.(CSEA)on paracetamol(APAP)-induced hepatic injury and its antioxidant activity in vitro.Methods Twenty KM mice were randomly divided into control group,model group,and CSEA low and high dose groups(50 and 200 mg·kg-1).7 days of after the administration,APAP(300 mg/kg)was injected intraperitoneally to establish a liver injury model.Detection of serum ALT,AST activity and oxidative stress indicators T-SOD,MDA,GSH and T-AOC contents;TNF-α,IL-6,and IL-1βin liver tissues were detected by ELISA assay;hepatic pathological changes were observed with HE staining,DPPH free radical and hydroxyl free radical scavenging experiments were performed to detect the in vitro antioxidant activity of CSEA.Results CSEA can significantly reduce ALT,AST,MDA,TNF-α,IL-1βand IL-6 levels,increase T-SOD,GSH and T-AOC levels and improve pathological changes of the liver tissues;there was strong scavenging ability on two different types of free radicals of DPPH and hydroxyl free radicals.Conclusion There was a good anti-oxidative capacity in vitro in the ethyl acetate fraction of Capparis spinosal L.and APAP-induced liver injury model,which may be related to antiqxidation and anti-inflammatory effects.
