哈蟆油蛋白对去势大鼠骨组织形态保护的实验研究 被引量:2

Experimental Study on the Morphological Protection of Bran Oil Protein in Ovariectomized Rats

摘要 目的观察大鼠骨密度(BMD)、生物载荷量及股骨组织切片形态学变化,评价哈蟆油蛋白对去势大鼠骨形态的保护作用。方法利用动物骨密度仪和小动物骨骼强度测定仪测量大鼠BMD和生物载荷量;采用HE染色法观察大鼠股骨骨小梁数量变化,应用免疫组化技术观察哈蟆油蛋白对ALP和BMP2表达的调节作用。结果与模型组比较,哈蟆油蛋白明显提高了大鼠的BMD(P<0.05),显著改善了股骨的最大载荷量(P<0.01);150mg·kg^(-1)哈蟆油给药组的骨小梁的排列更加有序,结构更加完整;同时低剂量和高剂量组的ALP和BMP2呈现阳性表达。结论哈蟆油蛋白能增强骨强度、降低骨脆性骨折风险,同时能保护骨小梁结构,促进ALP和BMP2的阳性表达,故哈蟆油蛋白对骨组织形态发挥一定的保护作用,可能参与骨代谢平衡,发挥防治绝经后骨质疏松的作用。 Objective To observe the changes of bone mineral density(BMD),biological load and morphological changes of femur tissue in rats,and to evaluate the protective effect of hare oil protein on bone morphology of ovariectomized rats.Methods BMD and biologic load of rats were measured by using animal bone densitometer and small animal bone strength tester.The number of femoral trabecular bone in rats was observed by HE staining.The immunohistochemical technique was used to observe the expression of haploid oil protein and ALP.Regulation of BMP2 expression.Results Compared with the model group,B.oleifera protein significantly increased BMD in rats(P<0.05),significantly improved the maximum femur load(P<0.01),and 150 mg/kg B.oleoresin administration group of bones.The arrangement of trabeculae is more orderly and the structure is more complete;at the same time,the ALP and BMP2 in the low-dose and high-dose groups show positive expression.Conclusion The hare oil protein can enhance the bone strength,reduce the risk of bone fragility fractures,at the same time can protect the trabecular structure,and promote the positive expression of ALP and BMP2.Therefore,haploid oil protein plays a certain protective effect on the bone tissue morphology and may participate in Balanced bone metabolism plays a role in preventing and treating postmenopausal osteoporosis.
