枸杞多糖通过抗氧化及抗凋亡作用对过氧化氢致血管内皮细胞损伤的保护作用研究 被引量:16

摘要 目的探讨枸杞多糖(Lycium barbarum polysaccharide,LBP)对氧化应激致大鼠主动脉血管内皮细胞(Rat Artery Endothelial Cells,RAEC)损伤的抗氧化及抗凋亡的保护作用,为进一步开发LBP的药用价值提供实验及理论依据。方法培养RAEC并随机分为3组:对照组、过氧化氢损伤组(H2O2组)、H2O2+LBP (440μg/ml)组,采用比色法检测培养细胞上清液中超氧化物歧化酶(SOD)活力、丙二醛(MDA)及NO(一氧化氮)水平;Western blot法检测细胞Bcl-2及Bax的表达水平。结果与对照组比较,H2O2组SOD活力与NO含量明显降低(P<0.05)、MDA含量明显升高(P<0.05);与H2O2组比较,H2O2+LBP组SOD活力及NO含量升高(P<0.05)、MDA含量降低(P<0.05)。与正常对照组比较,H2O2组Bcl-2表达下降,Bax表达升高,Bcl-2/Bax下降(P<0.05);与H2O2组比较,H2O2+LBP组Bcl-2蛋白的表达升高、 Bax表达下降及Bcl-2/Bax升高(P<0.05)。结论枸杞多糖可通过抗氧化和抗凋亡作用发挥对过氧化氢致主动脉血管内皮细胞损伤的保护作用。 Objective To investigate the effects of Lycium barbarum polysaccharide(LBP)on the oxidative stress rat aortic endothelial cells(RAEC)injury of the protection through antioxidant and antiapoptotic effect,for the further development of medicinal value of LBP provide experimental and theoretical basis.Methods RAEC was cultured and randomly divided into 3 groups:control group,hydrogen peroxide injury group(H2O2 group),H2O2+LBP(440 ug/ml)group.SOD activity,malondialdehyde(MDA)and nitric oxide(NO)levels in the supernatant solution of cultured cells were detected by colorimetry.The expression levels of bcl-2 and Bax were detected by Western blot.Results Compared with the control group,SOD activity and NO content in the H2O2 group were significantly reduced(P<0.05)and MDA content was significantly increased(P<0.05).Compared with the H2O2 group,the SOD activity and NO content in the H2O2+LBP group increased(P<0.05)and MDA content decreased(P<0.05).Compared with the normal control group,bcl-2 expression decreased in the H2O2 group,Bax expression increased,and bcl-2/Bax decreased(P<0.05).Compared with the H2O2 group,the expression of bcl-2 protein in the H2O2+LBP group increased,Bax expression decreased,and bcl-2/Bax increased(P<0.05).Conclusion LBP can play a protective role in the injury of aortic endothelial cells induced by hydrogen peroxide through its antioxidant and anti-apoptotic effects.
