摘要
目的研究紫地榆中4种成分对致龋菌细胞内乳酸脱氢酶和蔗糖酶的影响。方法以3种致龋菌(变形链球菌、粘性放线菌、嗜酸乳杆菌)为供试菌,测定不同浓度含药培养液对3种致龋菌培养液中乳酸脱氢酶和蔗糖酶的影响。结果除鞣花酸作用于嗜酸乳杆菌外,4种成分对3种致龋菌中两种酶均有抑制作用。其中对3种致龋菌细胞内乳酸脱氢酶的活性抑制作用最强的分别是没食子酸2 mg/mL(P<0.05)、儿茶素0.25 mg/mL(P<0.05)、没食子酸甲酯1 mg/mL(P<0.05)。对3种致龋菌细胞内蔗糖酶的活性抑制作用最强的分别是没食子酸甲酯2 mg/mL(P<0.05)、鞣花酸0.25 mg/mL(P<0.05)、没食子酸2 mg/mL(P<0.05)。结论紫地榆4种成分作用于致龋菌后使致龋菌中乳酸脱氢酶和蔗糖酶活性明显降低,可减少龋齿的发生。
Objective To study the effects of four components in the purple radix on lactate dehydrogenase and sucrase in cariogenic cells.Methods 3 kinds of cariogenic bacteria(Streptococcus mutans,Viscous actinomycetes and Lactobacillus acidophilus)were used as test bacteria to determine the effects of different concentrations of drug-containing culture solutions on lactate dehydrogenase and sucrase in three cariogenic broth cultures.Results In addition to ellagic acid acting on Lactobacillus acidophilus,the four components inhibited both of the three cariogenic bacteria.Among them,the strongest inhibitory effect on the activity of lactate dehydrogenase in three kinds of cariogenic bacteria was gallic acid 2 mg/mL(P<0.05),catechin 0.25 mg/mL(P<0.05),gallic acid A.Ester 1 mg/mL(P<0.05).The most inhibitory effects on the activity of sucrase in the three cariogenic bacteria were methyl gallate 2 mg/mL(P<0.05),ellagic acid 0.25 mg/mL(P<0.05),gallic acid 2 mg./mL(P<0.05).Conclusion The four components of the purple scorpion act on the cariogenic bacteria,which can significantly reduce the activities of lactate dehydrogenase and sucrase in cariogenic bacteria,which can reduce the occurrence of dental caries.
