摘要
目的基于网络药理学探讨葛根素注射液对高血压的作用机制。方法利用CTD数据库筛选葛根素注射液中的作用靶点,构建葛根素注射液作用靶点的蛋白互作网络;基于Genecards数据库筛选高血压相关的靶点,并利用venn工具绘制葛根素注射液药物靶点与高血压疾病靶点的韦恩图,并通过生物信息学手段对葛根素注射液对高血压的作用靶点进行GO分析及KEGG分析。结果经CTD筛选99个靶点,包括与高血压相关30个作用靶点;经Bonferroni校正P值均<0.01,经Kappa算法分为活性氧代谢过程、外分泌细胞凋亡、骨髓白细胞分化、细胞对外界刺激的反应等27大类别;得出19条信号通路,经Bonferroni校正P值均<0.01。结论研究结果初步预测了葛根素注射液对高血压的基本药理基础及功效机制,与现代药理学研究以及临床用药规律有一定契合,葛根素注射液治疗高血压具有多基因参与的特点,可能与细胞分化凋亡以及活性氧代谢有关。
Objective To explore the mechanism of Puerarin Injection on hypertension based on network pharmacology.Methods CTD database was used to screen the target of puerarin injection and construct the protein interaction network of the target of Puerarin injection.Genecards database was used to select the target of hypertension,and Venn tool was used to draw the Venn diagram of the target of puerarin injection and the target of hypertension.GO analysis and KEGG analysis of the target of Puerarin Injection on hypertension were performed by bioinformatics means.Results 99 targets were screened by CTD,including 30 targets related to hypertension.The P values were<0.01 corrected by Bonferroni,and were divided into 27 categories by Kappa algorithm:reactive oxygen metabolism,exocrine cell apoptosis,bone marrow leukocyte differentiation,and cell response to external stimuli.19 signal pathways were obtained,corrected by Bonferroni and the P values were<0.01.Conclusion The results of this study preliminarily predicted the basic pharmacological basis and efficacy mechanism of Puerarin Injection on hypertension,which was consistent with modern pharmacological research and clinical medication rules.Puerarin injection in the treatment of hypertension has the characteristics of multi-gene participation,which may be related to cell differentiation and apoptosis and reactive oxygen metabolism.
