苍术炒焦前后苍术苷A的含量比较研究 被引量:10

Comparison of the Content about Atractyloside A Before and After Atractylodes Rhizoma Being Deep-fried

摘要 目的 建立苍术饮片中苍术苷A的含量测定方法,对苍术炒焦前后苍术苷A含量进行比较研究。方法 采用高效液相色谱法(HPLC-ELSD)对苍术炒焦前后的色谱图进行比较,经与对照品比对,确定苍术苷A色谱峰位置,并测定苍术炒焦前后苍术苷A含量;色谱条件为Agilent C18(5μm,4.6 mm×250 mm)色谱柱,流动相(乙腈-水)梯度洗脱;柱温30℃,ELSD检测器,漂移管温度80℃,雾化气体为氮气,压力3.0×105 Pa;流速1.6 mL·min-1。结果 苍术苷A进样量在0.1012~2.0240μg范围内线性关系良好(r=0.9991),加样回收率在98.1%~98.9%区间;苍术炒焦后苍术苷A含量增幅在58.44%~87.00%。结论 焦苍术中苍术苷A含量明显增加,为苍术炒焦后的主要增量成分之一,可能是导致苍术炒焦药效改变的主要物质基础。 Objective To establish a method to determine the content of Atractyloside A in the slices of Atractylodis Rhizoma,and to compare the content of atractyloside A before and after Atractylodes Rhizoma being deep-fried.Methods HPLC-ELSD was used to compare the chromatogram of Atractylodis Rhizoma before and after being deep-fried,and compared with the chromatogram of reference substances for Atractyloside A.Then the chromatographic peak of Atractyloside A was determined,and their contents were determined in slices of Atractylodis Rhizoma before and after being deep-fried.The chromatographic conditions included:An analytical C18 column(Agilent,4.6 mm x 250 mm,5μm)and the mobile phase(acetonitrile-water)was gradient elution;the column temperature was 30℃,the drift tube temperature for the ELSD was set at 80℃,and the atomizating gas was nitrogen with the pressure of 3.0×105 Pa;The flow rate was 1.6 ml·min-1.Results The calibration curve of Atractyloside A was linear in the ranges of 0.1012~2.0240μg(r=0.9991),the sample recovery rate was in the ranges of 98.1%~98.9%;The content of Atractyloside A increased from 58.44%to 87.00%after the slices of Atractylodis Rhizoma being deep-fried.Conclusion The content of atractyloside A in the slices of Atractylodis Rhizoma was obviously increased,it is one of the main incremental components after being Fried and may be the main material basis for the change of efficacy of the focal Atractylodis Rhizoma.
