双黄益骨方对绝经期糖尿病骨质疏松症预防作用的实验研究

Experimental Studies on Preventive Effect of Shuanghuang Yigu Prescription on Menopausal Diabetic Osteoporosis

观察双黄益骨方对绝经期糖尿病骨质疏松症的预防作用。方法 采用双侧卵巢切除和链脲佐菌素腹腔内注射复合造模法,建立绝经期糖尿病骨质疏松症(MDO)的大鼠模型,设中药预防组、模型对照组及正常对照(伪手术)组,治疗6周后,检测脱氧吡啶啉(Dpd,为Dpd与尿肌酐的比值)、血清骨钙素(BGP)及左后肢股骨、腰椎(L_4,L_5)生物力学等指标:结果 尿Dpd、血清BGP中药预防组均明显升高,与正常对照组和模型对照组比较,均有显著性差异(P<0.01)。L_4椎体凹入试验,模型对照组、中药预防组与正常对照组比较,均具有显著性差异(P<0.05);中药预防组与模型对照组比较均值虽有所升高,但其差异无统计学意义。结论 双黄益骨方能刺激绝经期糖尿病骨质疏松症模型大鼠成骨细胞分泌,促进骨胶原的合成;降低破骨细胞活性,抑制骨吸收;从而可能改善MDO大鼠的骨质量,预防绝经期糖尿病骨质疏松症。

Objective To investigate the preventive effect of Shuanghuang Yigu Prescription (SYP) on menopausal diabetic osteoporosis (MDO). Methods Rat models of MDO were established by bilateral ovariotomy and intraperitoneal injection of STZ. Rats were divided into three groups: SYP group, model group and mimic operation group. After six - week treatment, urine desoxypyridoxine (DPD), blood B -glycerophosphatase (BGP) and biomechanics indexes of L_4, L_5 and left thighbone were measured. Results Urine DPD and blood BGP were obviously increased in SYP group( P<0.01 as compared with mimic operation group and model group). The difference of L_4 vertebral centrum cave test in the three groups were significant (P<0.05) but the increase of biomechanics indexes were insignificant between SYP group and model group. Conclusion SYP can improve the bone metabolism to prevent and treat MDO by simulating the secretion of osteoblast, promoting the formation of ossein, decreasing the activity of osteoclast and inhibiting the absorption of bone in rats with MDO.
