摘要
在测定了BmNPV-Ch(中国株)和HaMNPVvp39基因序列的基础上,推导出相应的氨基酸顺序.并与AcMNPV、OpMNPV、LdMNPV、BmNPV-Ja(日本株)的VP39蛋白的氨基酸顺序进行了比较.分析结果表明:在已知的各种杆状病毒中,VP39蛋白是一个保守性较强的蛋白质.Bm-NPV-ChVP39蛋白与AcMNPV、OpMNPV、LdMNPV、BmNPV-Ja和HaMNPVVP39蛋白的氨基酸同源性分别是93.7%、58.2%、39.9%、97.1%、92.8%.HaMNPVVP39蛋白与AcMNPV、OpMNPV、LdMNPV、BmNPV-Ja和BmNPV-ChVP39蛋白的氨基酸同源性分别为97.3%、59.0%、40.2%、93.1%、92.8%.通过氨基酸亲水性分析比较,探讨了VP39蛋白结构与杆状病毒进化间的关系.
The nucleocapsid protein gene (vp39) of Bombyx mori nuclear polyhedrosis virus (Chinese isolate, BmNPVCh) is sequenced which contains an open reading frame of 1 050bp long and codes for a polypeptide with a putative relative molecular weight (Mw) of 39 000 daltons consisting of 350 amino acid residues. The deduced amino acid sequence is compared with other VP39 amino acid sequences from 5 insect viruses (AcMNPV, OpMNPV, LdMNPV, BmNPVJa and HaMNPV). The Maximum homology of amino acid sequences and the conserved structural regions are analyzed with PROSIS. The results indicate that the main part of the secondary structure of the VP39 protein is a rather conservative structure containing high proportion of βsheets and βturn units; and that the homology of amino acid sequences are 93.7%, 58.2%, 39.9%, 97.1% and 92.8% respectively. The relationship of evolution about 6 insect viruses is explored.
