摘要
目的 探讨西布曲明 (MR)的减肥作用。方法 给体重为 2 0~ 2 4 g的♂小鼠腹腔注射金硫葡萄糖 (GTG) 80 0mg·kg-1一次以诱导产生肥胖 ,对照组动物则给以等体积的生理盐水 ;在造型后 2 1d ,将肥胖动物按体重随机均分为 5组 ,分别灌胃MR 3 0 ,6 0和 12 0mg·kg-1;安菲拉酮 90mg·kg-1,模型组和对照组则均给 0 5 %CMC 10ml·kg-1,1d 1次 ,连续 18d。末次给药后 2 4h ,动物同时禁食过夜后 ,将动物轻度麻醉后测其体长及体重 ,眼眶取血测定其血清甘油三酯 (TG) ,胆固醇 (TC) ,胰岛素和血糖浓度 ,取腹部、肾周围及生殖器周围白色脂肪组织称重 ,同时取肩胛下棕色脂肪组织称重 ;取生殖器周围白色脂肪组织作脂肪细胞记数。结果 MR各剂量组及阳性对照组均可使GTG小鼠的体重增加减少、使GTG小鼠的脂肪垫重量和Lees指数均减少、使肥胖动物的胰岛素和TG含量降低、使GTG小鼠每克脂肪组织的脂肪细胞个数增加 ,其表面积和体积均减少 ,与模型组比较差异有统计学意义。
AIM To assess the effect of sibutramine on obese mice. METHODS Male mice were induced obesity by IP gold thioglucose(GTG) 800 mg·kg -1 once,while the control mice were given the same valume saline.On the 21th day,the obese animals were devided into five groups and administraed orally sibutramine 3,6,12 mg·kg -1 or amfepramoni hydrochloridi 90 mg·kg -1 or 0 5% CMC for 18 days respectively. RESULTS In sibutramine treated GTG mice,the weight gain,weight of fat pads,Lee's indexes,size of adipocytes, the levels of serum TG and insulin were all reduced. CONCLUSION Sibutramine has an anti obesity action in GTG mice.
