K-ras基因转染鼠肺纤维细胞粘附分子的表达及临床意义

摘要 目的 :探讨细胞粘附分子在肿瘤细胞浸润转移中的意义。方法 :利用免疫荧光标记及聚丙烯酰胺凝胶电泳和Westernblot观察K ras基因转染的鼠肺 (KTRL)纤维细胞及正常鼠肺 (NRL)纤维细胞钙粘素 链蛋白复合体分子的表达。结果 :KTRL纤维细胞粘附分子表达明显异常 ,肌蛋白分布方式发生改变 ,粘附能力明显下降。结论 :KTRL纤维细胞作为肿瘤细胞的早期阶段 ,其粘附能力的改变 。 Objective: Our aim was to study the function of adhesive molecule in the process of tumor cell invasion. Methods: We studied lung fibrocytes in K-ras transformed rats and normal rats. The expression of cadherin-catenin complex was observed with immunofluorescence labeling technique, polyacrylamide gel electrophoresis, and western blot. Results: Obviously abnormalities was found in the expression of adhesive molecule of lung fibrocytes in K-ras transformed rats. The distribution of actin changed with significance decrease of adhesion. Conclusion: The change of cell adhesion in early stage of tumor may be a major factor for cell invasion and metastasis in advanced stage.
