摘要
目的 :研究奥曲肽在大鼠急性坏死性胰腺炎 (ANP)模型中对核因子κB (NF κB)活化的作用。方法 :大鼠ANP模型 ,随机分成 3组 ,对照组 ,ANP组 ,奥曲肽组。观察血清淀粉酶、一氧化氮、胰腺病理的变化及NF κBp6 5mRNA在胰腺组织的表达。结果 :应用奥曲肽后 ,奥曲肽组与ANP组比较 ,血清淀粉酶、一氧化氮明显下降 ,分别由 (8131.5 0± 883.97)U/L ,(81.4 7± 10 .6 7) μmol/L降至 (396 8.30± 993.91)U/L ,(6 9.89± 11.87) μmol/L ;胰腺损伤减轻 ;NF κBp6 5mRNA表达下降。结论 :奥曲肽抑制ANP大鼠NF κB的活化 ,并通过抑制NF κB的活化抑制诱导型一氧化氮合酶 (iNOS)的表达 ,改善大鼠ANP。
Objective: Our aim was to assess the effect of octreotide on nuclear factor-kappa B (NF-κB) activation in rats with acute necrotizing pancreatitis (ANP). Methods: Rat models with ANP were diivided into 3 groups: control ANP, and octreotide groups. The changes of serum amylase, nitric oxide (NO), pancreatic morphological damage, and the expressions of NF-κB p65 subunit mRNA were determined. Results: Levels of serum amylase and NO decreased significantly in ANP group compared with octreotide group [(8131.50±883.97)U/L vs(3968.30±993.91)U/L, P<0.05; (81.47±10.67)μmol/L vs(69.89±11.87)μmol/L, P<0.05 respectively]. Expression of NF-κB p65 subunit mRNA in octreotide group was lower than that in ANP group. Octreotide could alleviate pancreas destruction. Conclusion: Correlation between NF-κB activation, serum amylase, NO, tissue damage suggests a key role of NF-κB in the pathogensis of acute pancreatitis. Octreotide can inhibit the activation of NF-κB in ANP rats, thus inhibit the expression of iNOS and reverse the pancreatic damage of ANP rats.
