摘要
目的 :研究 genistein对兔晶体上皮细胞增殖及胞膜酪氨酸蛋白激酶 (TPK)、蛋白激酶C(PKC)活性的影响 ,探索使用 genistein防治后发性白内障。 方法 :兔晶体上皮细胞培养 ,应用Casnetllie改良法检测不同浓度genistein及bFGF作用后 ,兔晶体上皮细胞膜TPK活性的变化 ;同位素掺入法检测 genistein及bFGF作用后 ,兔晶体上皮细胞胞膜及胞浆PKC活性的变化。结果 :不同浓度 genistein作用后兔晶体上皮细胞TPK活性均较对照组明显下降 ,genistein可抑制bFGF诱导的TPK活性升高 ,genistein对PKC活性无明显抑制作用 ,但可抑制bFGF诱导的PKC活性升高。结论
Objective: Our aim was to assess the effect of genistein on tyrosine protein kinase and protein kinase C (PKC) activation of rabbit lens epithelial cells (RLEC). Methods: The RLEC was cultured in vitro. Isotope incorporation method was used to measure activities of tyrosine protein kinase and protein kinase C in cell membrane. Results: The activation of tyrosine protein kinase was significantly inhibited by genistein. Activation of PKC in cytoplasm of lens epithelial cells induced by bFGF could be decreased by genistein. Conclusion: Genistein may be effective for the prevention of after cataract.
