摘要
目的化学合成AP-N抑制剂类天然抗癌化合物AHPA-Val甲酯,初步探讨其抗癌作用与免疫学活性的关系。方法以丙烯酸、苯甲醛和缬氨酸甲酯为起始原料,经氯代、硝化、消除、加成、缩合和催化氢化等7步反应合成了AHPA-Val甲酯,用荷S180的昆明系小鼠进行初步抑瘤活性试验和增强免疫功能试验。结果AHPA—Val甲酯的化学结构经IR和1H—NMR波谱解析和元素分析所证实。在20 mg/(kg·d)剂量下,对小鼠S180的抑瘤率为28.3%,免疫学廓清指数与对照组相比有显著性差异(P<0.05)。结论AHPA—Val甲酯的抗癌活性与其对机体的免疫增强作用有关。
Aim To synthesize the AHPA-Val methyl ester of an AP-N inhibitor, a natural anticancer compound and explore initially the relationship between the anticancer and immunology activity. Method AHPA-Val methyl ester was synthesized through chlorination, nitration, elimination, addition, condensation and catalytic hydrogenation from acrylic acid, benzaldehyde and methyl valate.The initial tests of the antitumor and immunology activity were conducted in mice planted S180. Results The chemical structrue of AHPA-Val methyl ester was identified by IR, 1H-NMR and elemental analysis. The antitumor rate is 28.3% against mice S180 at the dosage of 20 mg/(kg·d). The immunophagocytic index shows significant effect compared with that of the control group(P<0.05).Conclusion The anticancer activity of AHPA-Val methyl ester is related to the immunopotentiation in the body.
