CD30^+间变性大细胞淋巴瘤细胞系的建立及其生物学特性

Establishment of a Human CD30^+ Anaplastic Large Cell Lymphoma Cell Line and Its Biological Characteristics

摘要 由 1例非霍奇金淋巴瘤患者的胸水标本通过液体培养法建立了 1株CD3 0 + 间变性大细胞淋巴瘤细胞系 ,并对其生物学特性进行研究。所建立的细胞系可以在含 10 %小牛血清的RPMI 1640培养液中持久增殖 ,倍增时间约为 3 6小时 ,目前已传代 12 0余次 ,生长良好 ,暂命名为SH 1。光镜下细胞形态与典型的CD3 0 + 间变性大细胞淋巴瘤 (anaplasticlargecelllymphoma ,ALCL)细胞的形态十分类似 ,电镜下可观察到胞浆内存在较多病毒颗粒。细胞组织化学染色 :光镜下过氧化物酶POX阴性 ,糖原染色PAS阳性 ,酸性磷酸酶 (ACP)呈强阳性 ;电镜下髓过氧化物酶 (MPO )阴性 ,血小板过氧化物酶 (PPO)阴性。EB病毒核心抗原 1(EBNA 1)呈阳性。细胞免疫表型为CD3 0 + 、CD45+ 、HLA DR+ ,而EMA、CD3 4、CD3 8、CD2、CD3、CD4、CD7、CD8、CD10、CD15、CD19、CD2 0均呈阴性。染色体分析示SH 1细胞系核型为二倍体或四倍体核型 ,并具有明显的结构和数量异常 ,但未见典型t(2 ;5)。结论 :所建立的细胞系为CD3 0 + ALCL细胞系。 A CD30 + anaplastic large cell lymphoma (ALCL) cell line wa s established from the mononuclear cells isolated from pleural effusion of a pat ient with non-Hodgkin′s lymphoma. The cell line′s biological characteristics were analyzed. The results showed that t he established cell line could survive and proliferate in RPIM 1640 medium; the Wright-Giemsa-stained cells were exactly similar to malignant cells of CD30 + ALCL in morphology, with many diffuse virus granules in cytoplasm; the cytochemical staining of the cells sho wed the following reactivity pattern: positive for acid phosphatase (ACP) and pe riodic acid-Schiff (PAS), negative for peroxidase (POX), myeloperox idase (MPO) and platelet peroxidase (PPO). The immunoprofile of the cells was po sitive for CD45, HLA-DR, CD30 and negative for EMA, CD34, CD38, CD2, CD3, CD4, CD7, CD8, CD10, CD 15, CD19 and CD20. The cytogenetic analysis showed complicated qualitative and quantitative abnormality of chromosomes, without typical t(2;5). It is conc luded that the established cell line is CD30 + anaplastic large cell lymphoma c ell line.
