摘要
目的 观察碱性成纤维细胞生长因子的单克隆抗体 b FGF- MAb对人卵巢癌细胞的增殖、卵巢癌腹腔移植瘤裸鼠的生存率和肿瘤血管生成的影响 ,为卵巢癌的临床生物治疗提供实验基础。方法 1将人卵巢癌细胞株 SKOV3接种于 2 4孔板 ,加入不同浓度的 b FGF- MAb,每日行结晶紫染色、Vector2 - 14 2 0多标记计数仪比色 ,测定 OD4 90 值 ;2将 SKOV3细胞接种于 BAL B/ c裸鼠腹腔形成卵巢癌腹腔移植瘤模型 ,每周 2次将 b FGF- MAb注射于腹腔 ,比较裸鼠生存时间 ;3解剖裸鼠 ,计数肠系膜上种植瘤的个数并称重 ;4对移植瘤组织进行 CD31 的免疫组化染色后测定肿瘤内微血管密度 (MVD)。结果 1b FGF- MAb在 0~ 15 μg/ ml的范围内 ,其抑制 SKOV3细胞增殖的作用呈浓度依赖性 ,实验第 4 d,15 μg/ m l组能抑制细胞增殖的 2 4 % ;2腹腔注射 b FGF- MAb的裸鼠生存时间较对照组平均延长 10 d;3b FGF- MAb组肠系膜上肿瘤种植灶的数目和重量分别是对照组的 70 .6 %和 6 9.2 % ;4 b FGF- MAb组腹腔移植瘤 MVD是对照组的 6 2 .8%。结论 b FGF- MAb能明显抑制卵巢癌的生长和肿瘤血管生成 。
Objective To investigate whether the monoclonal antibody of basic fibroblast growth factor (bFGF MAb) inhibits the proliferation, invasion and angiogenesis of ovarian cancer. Methods ①Human ovarian cancer cells SKOV 3 were planted in 24 well dishes, to which was added bFGF MAb at different concentrations, and crystal violet staining was performed daily for 8 days, then cell numbers were counted by OD 490 determination. ②SKOV 3 were transplanted intraperitoneally to BALB/c nude mice. One week later, bFGF MAb was injected intraperitoneally twice a week, and the survival time of nude mice, the number and weight of the metastatic tumors on the mensentery were observed or measured. ③Intratumoral microvessel density(MVD) was measured by immunohistochemical staining for CD 31 . Results ①bFGF MAb inhibited the proliferation of SKOV 3 in the concentration dependent manner ( P <0.05). ②The average survival time of nude mice in bFGF MAb group increased by 10 days over that of control. ③The average number and weight of metastatic tumors on mensentery in bFGF MAb group constituted 70.6% and 69.2% of those in control group, respectively. ④The MVD in bFGF MAb group accounted for 62.8% of the MVD in control group. Conclusion bFGF MAb can inhibit the proliferation and angiogenesis of ovarian cancer markedly, so it promises to have application in the biological treatment of ovarian cancer.
