摘要
目的 研究氧化应激反应中氟对血管平滑肌细胞毒性中的作用机制。方法 对大鼠胸主动脉平滑肌细胞采用组织块培养法。应用生化方法检测细胞内蛋白和丙二醛 (MDA)含量及细胞超氧化物歧化酶 (SOD)和过氧化氢酶(CAT)的酶活性。结果 随着氟作用时间的延长和氟浓度的增加 ,细胞内蛋白含量明显下降 ,在同一时间段MDA含量随着投氟浓度的增加而增加 ,72h时间段的MDA含量较 2 4h时间段的相同氟作用浓度下MDA明显下降 ,抗氧化酶SOD、CAT的活性在 72h时间段比 2 4、48h时间段的相同氟浓度组活性明显增高。结论 随着氟作用时间的延长 ,浓度的增加对血管平滑肌细胞有明显毒性 。
Objective To study the mechanism of oxidative stress in fluoride treating vascular smooth muscle cell. Methods Utilizing biochemical methods to test the level of vascular smooth muscle cell protein and MDA ,the activity of SOD and CAT. Results The concentration of protein is lower when the longer time and the higher content fluoride treated the vascular smooth muscle cell. The MDA showed lower in the vascular smooth muscle cells treated by fluoride 72 hour than that of 24 hour. The activity of SOD and CAT obviously higher in vascular smooth muscle cells treated by fluoride 72 hour than that of 24 hour and 48 hour.Conclusions Fluoride did obvious harm to vascular smooth muscle cells while its content and time increased; oxidative stress possibly gets involved in the toxic mechanism of fluoride.
