摘要
目的 :探讨白细胞介素 - 13(IL - 13)对肾小球系膜细胞 (MC)增殖及其细胞周期的影响。方法 :用甲基偶氮唑法 (MTT)测定MC增殖 ,流式细胞术分析细胞周期。结果 :IL - 13在 1ng/ml、5ng/ml、10ng/ml、10 0ng/ml浓度范围呈剂量依赖性抑制 5 %FCS培养的和脂多糖 (LPS)诱导的MC增殖 ,抑制率分别为 19.8%和 2 9.2 3%、2 4 .35 %和37.0 5 %、30 .0 3%和 4 6 .6 %、4 6 .93%和 5 5 .2 3%。IL - 13作用MC 4 8h ,使MC较多滞留于G1期 ,而S期细胞减少。结论 :IL -
Objective:To study the effects of interleukin-13(IL-13)on proliferation and cell cycle of rat mesangial cells.Methods:Cell prolifration was measured by MTT method.Flow cytometry was used to analyse cell cycle distribution.Results:At the concentration of 1 ng/ml,5 ng/ml,10 ng/ml and 100 ng/ml,IL-13 inhibited the proliferation of 5% FCS-cultured and LPS-induced mesangial cells by 19.83% and 29.23%,24.35%and 37.05%,30.03%and 46.67%,46.93%and 55.53%,respectively.The percentage of mesangial cells in G 1 phase markedly increased,while the percentage of mesangial cells in S phase markedly decreased after 48 h incubation with IL-13.Conclusion:IL-13 inhibit proliferation of mesangial cells.The inhibitory mechanism is at least partially through regulating cell cycle.
