摘要
为检测酱油和醋中黄曲霉毒素的含量 ,建立了免疫亲和层析净化荧光光度法。试样由甲醇 -水提取 ,提取液被过滤、稀释后 ,上交联着黄曲霉毒素B1、B2 、G1、G2 特异抗体的免疫亲和层析柱净化。以甲醇通过免疫亲和层析柱洗脱 ,溴溶液衍生 ,荧光光度计测定衍生物的黄曲霉毒素 (B1+B2 +G1+G2 )含量。酱油和醋中检出限分别为 2 5 μg kg和1μg kg。添加回收率在 85 %以上。该方法准确、简单、快速、安全。
Aflatoxins in soy sauce and vinegar were determined by a fluorometric method after cleaned\|up by immunoaffinity column.The samples were extracted with CH\-2OH\|H\-2O and then filtered,diluted with water,and cleaned up by an immunoaffinity column containing monoclonal antibody specific for aflatoxins B\-1,B\-2,G\-1 and G\-2.After washing with CH\-2OH to remove aflatoxins from attached antibodies,the total aflatoxins (B\-1+B\-2+G\-1+G\-2) were reacted with bromine solution (SFB) and determined immediately by fluorometry.The detection limits for soy sauce and vinegar analysis reached 2 5 μg/kg and 1 0 μg/kg respectively.And the recoveries are more than 85%.This method is accurate,simple,rapid and safe.
