温肾健脑法对脑萎缩智能状态及老龄小鼠海马区超微结构的影响

Influence on Mental Status of Brain Atrophic Patients and Ultrastructure in Mice of Warming Kidney and Strengthen Brain

目的:观察温肾健脑法对脑萎缩患者智能状态的临床疗效及对老龄小鼠海马区超微结构的影响。方法:采用随机分组、阳性药物对照方法,治疗组用温肾健脑法组方的益肾健脑颗粒2号,临床对照组用吡拉西坦片,动物实验对照组用维生素E。结果:中医证候总有效率治疗组95.0%,对照组60.0%,组间比较有统计学意义(P<0.01);智能状态的评定选用MMSE、HDS和ADL等量表,结果两组治疗前后自身比较均有统计学意义(P<0.01),组间比较均无统计学意义(P>0.05)。动物实验中发现老龄小鼠脑神经细胞结构正常,但出现海马CA1与CA3区神经细胞密度降低,线粒体老化,胞质中可见脂褐素颗粒和溶酶体等老化改变,而温肾健脑法和维生素E可延缓脑神经细胞的这种改变,从而为临床治疗脑萎缩提供了实验依据。结论:温肾健脑法能够改善脑萎缩患者的智能状态,并能延缓老龄小鼠海马区神经元的老化改变。

Objective: To explore the curative effect of Warming Kidney and Strengthen Brain on mental status of brain atrophic patients and its influence on ultrastructure in aged mice. Methods: Randomed the cases into two groups: the treatment group was treated with method of Warming Kidney and Strengthen Brain; and other subset patients were treated with Piracetam. In animal experiment, animals of the control group were treated with Vitamin E. Results: The effective rate of Chinese Medicine syndrome was 95.0% in treatment group and 60.0% in control group. There was significant difference in these two groups (P<0.01). Quantative Forms of MMSE,HDS and ADL were used to valued the mental status. All of these parameters were significantly ameliated in both of two groups' patients after treated, but there were no significantly different between these two groups. In animal experiment, the structure of nerve cell is normal in aged-mice, but the density of neuron was decreased in campus CA1 and CA3, and aged change of mitochondrion. The lipofuscin granule and aged change of lysoome, etc. can be found in neuron cytoplasm. All of these changes were ameliorated by the method of Warming Kidney and Strengthen Brain. Conclusion: The method of Warming Kidney and Strengthen Brain can ameliorate the mental status of brain atrophic patients and delay the aging change of in aged-mice.
