愈银方含药血清对角质形成细胞增殖与凋亡的影响

Regulatory Effects of Serum of Yuyin Recipe on Proliferation and Apoptosis of Kerationcytic Line

目的:探讨愈银方对角质形成细胞株COLO-16增殖及凋亡的调节作用。方法:利用体外细胞培养技术、MTT法、流式细胞仪、荧光显微镜技术检测愈银方的大鼠含药血清对COLO-16细胞株增殖及凋亡的影响。结果:愈银方含药血清对COLO-16细胞株增殖的抑制作用与浓度呈一定的关系,浓度越高则抑制作用越强。含药血清的浓度为10%、20%;设相应的正常大鼠血清浓度为对照组,培养48h后,COLO-16细胞的凋亡率分别为12.93%、19.80%,正常对照组为3.71%、10.96%,两者相比差异显著。结论:抑制COLO-16细胞株的增殖及诱导其凋亡可能是愈银方治疗银屑病的机理之一。

Objective:To study the effect of Yuyin Recipe on cultured keratincytic line colo-16 proliferation and apoptosis in vitro. Methods:The effect of serum of Yuyin Recipe on the proliferation of keratinocytic line colo-16 was observed using the cultured colo-16 in vitro and serum phamacological assay. Further studied the colo-16 apoptosis treated by Yuyin Recipe with fluorescence dying observation and flow cytometry assay. Result:MTT showed it could inhibit the proliferation of colo-16, their effects enhanced as concertraion increased and time prolonged. Also inducing apoptosis of colo-16 by flow cyctometry assay and fluorecence dying observation. Conclusion:Inhibition on over proliferration and inducing apoptosis of Kerratincytic line colo-16 may be one of the mechanisms of Yuyin Recipe in curing psoriasis.
