摘要
目的:探讨原代培养的人脑胶质瘤细胞是否分泌及表达白细胞介素6(Interleukin-6,IL-6),以及IL-6在人脑胶质瘤发病中的作用。方法:采用手术中直接获得的人脑胶质瘤组织,进行胶质瘤细胞的原代培养,并分别采用四唑盐比色分析法(MTT)及逆转录聚合酶链反应(RT-PCR)方法测定人脑胶质瘤细胞中IL-6的分泌及表达;再分别采用IL-6及IL-6单抗处理脑胶质瘤细胞。结果:①原代培养的人脑胶质瘤细胞可自发分泌IL-6,而正常人脑胶质细胞则无IL-6的分泌;②30例人脑胶质瘤标本中有25例表达IL-6基因,且其表达量与脑胶质瘤的恶性程度呈正相关关系;③IL-6能刺激脑胶质瘤细胞的增殖,而IL-6单抗则能抑制脑胶质瘤细胞的增殖。结论:IL-6在人脑胶质瘤中的异常分泌及表达可能是参与和维持脑胶质瘤恶性增殖的原因之一。
Objective:To explore the secretion and expression of interleukin-6(IL-6)in isolated fresh glioma cells,and to confirm the role of IL-6in the occurring and developing of human gliomas.Methods:The fresh isolated human glioma cells were cultured at different time,MTT and RT-PCR meth-ods were used to detect the biological activity of IL-6and the expression of IL-6gene in human glioma cells.Different concentrations of IL-6and IL-6McAb were used to act on fresh isolated human glioma cells.Results:①The isolated fresh human glioma cells secreted IL-6spontaneously without any stimulants.②The expression of IL-6gene was detectable in25tumor specimens(83.3%),and malignant tumors of high grade expressed significantly larger amount of IL-6gene compared to those of low grade.③IL-6stim-ulated but IL-6McAb suppressed the proliferation of human glioma cells.Conclusion:The abnormal se-cretion and expression of IL-6may play an important role in sustaining the malignant proliferation of hu-man gliomas.
