摘要
目的:应用电子显微镜观察白细胞介素-2、α-干扰素对体外培养成纤维细胞超微结构的影响,阐述白细胞介素-2、α-干扰素治疗瘢痕作用机制。方法:成纤维细胞培养后加入白细胞介素-2和α-干扰素,继续培养两三天,并做空白对照比较,通过扫描和透射电镜观察成纤维细胞微结构。结果:扫描电镜观察结果成纤维细胞表面纤维状物质和泡状突起明显减少。透射电镜观察结果成纤维细胞胞质内粗面内质网明显减少,所见内质网显著扩张,腔中内容物稀少,有的甚至雾泡化,粗面内质网上的核蛋白体有不同程度的脱颗粒和解聚现象,线粒体肿胀或空泡化。结论:白细胞介素-2、α-干扰素对成纤维细胞合成胶原有抑制作用。白细胞介素-2、α-干扰素抑制胶原生成可能是治疗瘢痕作用机制的一个方面。
AIM: To observe the effect of interleukin 2 and interferon α (IFN α ) on the ultrastructure of fibroblasts cultured in vitro and to explain the mechanism in treatment of scar. METHODS:Fibroblasts cultured in vitro Scanning were continuously cultured in interleukin 2 and IFN α for 2- 3 d. Scanning electron microscope (SEM) and transmission electron microscopy (TEM) were used to observe the ultrastructure of fibroblasts, and then the results were compared with the normal ones.RESULTS:SEM showed that fibrous material and alveolate process reduced markedly. TEM showed that the number of cytoplasmic rough endoplasmic reticula (RER) obviously reduced in fibroblasts, with endoplasmic reticulum expanding and rare content. Some even showed aerosol vacuoles. Different degrees of abscission and deploymerization were found in ribosome of RER with mitochondria swelling or vacuolating.
