摘要
目的:观察SD大鼠角朊细胞在体外培养的生长特性。方法:采用dispase酶消化法从SD大鼠仔鼠皮肤获得角朊细胞,在体外进行培养并传代,采用倒置显微镜和MTT法观察细胞的形态学变化和生长曲线。结果:通过酶消化法可以获得较纯的角朊细胞,原代培养10d细胞即可以达到融合,传代后细胞部分发生变性,呈成纤维细胞样,传代次数高,成纤维样细胞比例也相应增高。结论:SD大鼠角朊细胞可以在体外培养分裂增殖。
AIM:To study the characteristics of rat keratinocytes growth in vitro.METHODS:Keratinocytes harvested from rat skin by enzyme digestion were cultured in vitro.When the cells reached subconflunce,they were passaged with typsin.Phase microscope and cell growth curve were employed to observe the biological characteristics of keratinocytes.RESULTS:Simple keratinocytes can be harvested by dispase enzyme digestion.The primary cells showed superior proliferative ability and reached subconflunce(70) 10 days after seeded.The shape of some cells got changed and demonstrated the shape of fibroblast in the first passage.The proportion became higher in the second and third generation.
