摘要
目的 :观察L -精氨酸对高脂喂饲大鼠动脉核因子 -κB的合成影响。方法 :雄性Wistar大鼠 4 5只 ,随机分为NC组 (普通饲料及饮水 ) ,CC组 (高胆固醇饲料及饮水 ) ,AC组 (高胆固醇饲料喂饲的同时饮水中含 3%L -精氨酸 ) ;每组 15只 ,喂饲 12周后采动脉血 ,处死大鼠取腹主动脉全长利用WesternBlot检测核因子 -κB的含量。结果 :AC组及CC组大鼠血清胆固醇浓度无明显差异 ,但高于NC组 (P <0 .0 1) ,AC组血清一氧化氮水平明显高于CC组 (P <0 .0 1) ,AC组核因子 -κB的合成与NC组无差异 (P >0 .0 5 ) ,但低于CC组 (P <0 .0 1)。结论 :L -精氨酸抑制大鼠动脉核因子 -κB合成的作用可能是其抑制高脂膳食诱发的动脉粥样硬化的分子机制之一。
Objective:To investigate the effect of L-arginine on nuclear factor kappa B expression in the aorta of hypercholesterolemic rats.Method:Forty five male Wistar rats were randomized into three groups: Group NC was given normal diet ( NC, n=15), group CC hypercholesterolemic diet (CC, n=15) and group AC hypercholesterolemic diet supplemented with 3% L-arginine in drinking water (AC, n=15). After 12 weeks blood samples were collected for biochemical studies, and the aortas were harvested for Westem Blot.Results:Serum cholesterol concentrations of rats of the AC and CC groups were no significant difference ( P >0.05), but significantly higher than those of the NC group ( P <0.01). Serum nitric oxide concentrations of the NC and AC groups were no significant difference ( P >0.05), but significantly higher than those of the CC group ( P <0.01).Dietary L-arginine supplementation (group AC) significantly reduced the expression of nuclear factor kappa B ( P <0.01).Conclusions:Inhibitory effect of dietary L-arginine on nuclear factor kappa B expression may be one of the mechanisms of the anti-atherogenic effect of L-arginine.
