摘要
目的:研究和比较人晶状体上皮细胞(human lensepithelial cell,HLEC)在以聚甲基丙烯酸甲酯(polymethyl-methacrylate,PMMA)、硅凝胶(silicone,SI)和丙烯酸酯(acrylic,ACRY)为光学部材料的人工晶状体(intraocular lens,IOL)表面的粘附情况,以比较三种不同的人工晶状体材料的细胞粘附特性。方法:原代HLEC体外培养取材于角膜移植供体眼的晶状体。经过细胞鉴定后,将培养的第二代细胞以5×10~4/ml的密度接种到三种IOL表面培养。1d、3d、7d和14d后分别在倒置相差显微镜下观察细胞形态,并记数各种材料表面粘附的HLEC。记数结果进行方差分析。结果:细胞记数结果是,三种不同材料IOL表面粘附的HLEC数量不同:ACRY表面粘附的HLEC数量最多,1 d为(336.33±15.22)个/mm^2,3 d为(342.67±12.6)个/mm^2,7 d为(320±13.6)个/mm^2,14 d为(292±11.7)个/mm^2;SI最少,1 d为(10.67±2.26)个/mm^2,3 d为(6±3.92)个/mm^2,7 d为(2±1.57)个/mm^2,14d时细胞完全消失;PMMA界于两者之间,1d为(52±1.96)个/mm^2,3 d为(60.67±9.86)个/mm^2,7 d为(32.33±2.24)个/mm^2,14 d为(6.33±3.12)个/mm^2。不同材料表面粘附的细胞数量差异有显著统计学意义(P<0.01)。三种材料表面粘附的细胞形态亦有明显差异:ACRY表面的细胞伸展良好,2w后形态?
Objective: To evaluate the ability of human lens epithelial cells(HLECs) cultured in vitro to adhere to the surfaces of intraocular lenses composed of different materials. Poly(metbyl-methacrylate) (PMMA), silicone and bydrophobic acrylate (Acrysof) were used as testing materials. Methods: Primary culture of HLECs was established from lens anterior capsules obtained from donor eyeballs for comeal transplantation. HLECs were identified using polyantibody to α-crystalline. Cells in passage 2 were used for adhesion assay. 20 μL cells were seeded on the surface of three IOL materials at a cell density of 5×104/ml. Then cells were incubated with IOLs for 2 weeks. The mean number of cells that attached per square millimeter was calculated for each material after 1 day, 3 days, 1 week and 2 weeks. Meanwhile, the morphology of attached cells was observed under phase contrast microscopy. An analysis of variance was performed, with significance tested at the P = 0.01 level Results: After incubation for 1 day, the mean lumbers of attached HLECs on PMMA,silicone and acrylic were(52± 1.96)cells/mm2, (10.67±2. 26)cells/mm2 and (336.33±15. 22 ) cells/mm2, respectively. At 3 days, the average numbers of attached cells on the three materials mentioned above were(60.67±9.86)cells/mm2,(6±3.92)cells/mm2 and (342.67±12.6)cells/mm2, respectively. At 1 week, the mean numbers of HLEC on the three materials were(32.33±2.24 ) cells/mm2, (6±3.92) cells/mm2 and (320±13.6)cells/mm2, respectively. At 2 weeks, mean HLECs on PMMA and ACRY were(6.33±3.12)cells/mm2 and (292± 11.7)cells/mm2. No cells at all were observed on silicone. The difference in HLEC numbers on different IOL materials was statistically significance ( P < 0.01) . The morphology of cells attached to the three different materials was also different. HLECs spread well on ACRY and bad a regular shape. Even after incubation for 2 weeks,cells on ACRY still had the typical characteristics of epithelium and were firmly attached. HLECs on PMMA were only partially spread. They lost epithelial morphology and began their transformation to the fibroblast stage after 2 weeks. Those on silicone, which appeared to be small and round, didn't spread at all. They all disappeared within 2 weeks. Condition: The adherence of HLEC to IOLs was influenced by the surface property of IOL material , which may influence the occurrence of postoperative posterior capsular opacification after cataract surgery.
