摘要
目的 :研究脐血单个核细胞 (CB MNC)经脂多糖 (L PS)刺激后生成 IL- 15的动态变化。方法 :采用 EL ISA法对 14例 CB和 2 0例成人外周血 (APB) MNC经 L PS刺激后培养 6、12、2 4和 4 8h的上清进行测定。结果 :CB和 APBMNC经 L PS刺激后 ,培养 6 h的上清液中即可检测到 IL- 15蛋白 ,动态检测至培养的 4 8h,发现随培养时间的延长 ,其IL- 15的含量也不断增加 ,但脐血 MNC培养上清液中 IL- 15的含量明显低于成人外周血 (P值分别 <0 .0 5 ,0 .0 1,0 .0 1,0 .0 1)。结论 :CBMNC生成 IL- 15能力低下 ,在一定程度上反映了 CB的免疫特性 ,同 APB相比 ,CB免疫功能发育不成熟。
Objective:To investigate the dynamic change of interleukin-15(IL15) in cultured mononuclear cells(MNC) isolated from cord blood(CB) stimulated by lipopolysaccharide(LPS).Methods:The IL-15 of LPS-stimulated MNC culture supernatants at 6, 12, 24, 48 hours were determined by enzyme-linked immunosorbent assay (ELISA) in CB of 14 samples, and compared with adult peripheral blood(APB) of 20 samples.Results:IL-15 could be detected at 6 hours upon LPS- stimulation. The levels of IL-15 were gradually increased with increasing time. But the IL-15 levels in LPS-stimulated CB MNC were lower than that of APB MNC at each time point(P<0.05, 0.01, 0.01, 0.01, respectively).Conclusion:The reduced production of IL-15 from CB MNC in response to stimulation may contribute, in part, to the immaturity of CB immunity functions.
