摘要
利用载体pAM1146与烟草花叶病毒35S启动子,鲈鱼生长激素基因GH连接,构建携带鲈鱼生长激素基因的整合型表达载体pAMGH,用自然转化方法将pAMGH导入蓝藻7942,筛选到6株抗壮观霉素,氨苄青霉素敏感的藻落.提取抗性藻落的基因组总DNA,用生长激素基因特异引物进行PCR检测,结果证实鲈鱼生长激素基因已经整合进入蓝藻7942的染色体中.
By ligating vector pAM1146, tobacco mosaic virus promoter 35S and fish growth hormone gene GH,an integrative expression vector named as pAMGH was constructed.pAMGH was transformed into Synechococcus sp.pcc7942 by natural transforming method.6 spectinomycin resistant and ampicillin sensitive colonies were screened out.The total DNA of these colonies were extracted and PCR using specific primers for the GH gene was taken out for further analysis.4 colonies were confirmed to have GH gene integrated into the chromosome of cyanobacterium7942.
