摘要
对58例健康男女及2例有表型异常疑有染色体疾病者,分别按自定标准取头发毛囊10~15个,以常规直接法及改良法制备染色体标本;同时作30例外周血培养行染色体组型分析,以便相互对照.共分2组,A组30例为随意取材组,男11例,女19例,平均年龄28.83岁.B组30例,为选择取材组,男11例,女19例,平均年龄28岁.结果表明:①随意取材所获中期分裂相阳性例数占56.66%;选择取材占83.33%,B组较A组显著增高(P<0.025).②毛囊细胞制备染色体不同方法的比较证明,改良法具有明显优点,有非常显著意义(P<0.005).并就毛囊细胞快速制备染色体的理论依据及如何采集良好毛发标本等有关问题进行了讨论.
Rapid preparation of chromosomes specimens was carried out with humam hair follicle epithelial cells. Samples of 10~15 follicles each were taken from 58 healthy males and females and 2 persons with abnormal phenotype and suspected to have chromosomal desease. For the purpose of contrast, peripheral blood from 30 subjects was cultured to analyse karyotypes. The subjects were divided into two groups. Group A consisted of 30 persons that were chosen at random, with an average age of 28. 83 years. Group B included 30 persons chosen by selection, with an average age of 28 years. The result showed that the positive rate of mitotic metaphase was 56.66% in the group chosen at random and 83.33% in the group chosen by selection, showing a significant difference between the two groups. (P<0. 025). The comparison of different methods of chromosome preparation showed obvious advantages of the improved method and its remarkable significance (P<0. 005). Questions as to the theoretical basis of the rapid preparation of chromosomes with hair follicles and how to select good samples of hair, etc are discussed.
