斑点酶联免疫吸附试验诊断猪囊虫病的研究

STUDIES ON THE DIAGNOSIS OF CYSTICERCOSIS CELLULOSAE BY DOT-ELISA

通过Sephadex G-200层析提纯猪囊虫的囊液抗原和匀浆抗原,并以此抗原建立Dot-ELISA法来诊断猪囊虫病。该法在接到病料后1.5h内准确报告结果,结果判定不但简易直观,而且还可以作为技术资料长期保存。本法以硝酸纤维素膜作为载体,抗原用量、其他器材和试剂用量都大为减少,因而降低了检测成本。与四种寄生虫病(旋毛虫、弓形虫、细颈囊尾蚴、猪蛔虫)阳性血清不发生交叉反应。对按“四部”规程肉检确认为猪囊虫阴性血清42份,阳性血清17份,经 Dot-ELISA 法检测,其阴性符合率为95.24%,阳性符合率为100%,经三次重复,重复率为100%。对西宁市肉联厂的536份商品猪血清进行检测,其猪囊虫病阳性检出率为4.29%。试验结果证明,该法符合“简便、快速、敏感、准确、经济”的原则,尤其易于在基层推广应用。

Cyst fluid antigen and homogenate antigen of Cysticercus cellulosaewere purified by Sephadex G-200 chromatography.Dot-ELISA diagnosing cys-ticercosis cellulosae wae established by the antigen.This method can reportresult accurately after 1.5 hours of receiving pathogenic material.The re-sult can be judged simply and directly and preserved for a long time as thetechnical information.It was carried with nitric acid fibrinous membrance.The antigen dose,reagent dose and instruments are decreased conside-rably,therefore,the detective cost is very cheap.And it has no cross re-action with the positive sera of other four parasitosis:trichinosis,toxo-plasmosis,cysticercosis tenuicollis,ascariosis suis.42 negative sera and 17positive sera of C.cellulosae confirmed by the “four ministries” meat ex-amination rule were examined by Dot-ELISA,the coincidence rates ofnegative and positive respectively were 95.24% and 100N.The results of threerepeated examinations were completely coincident,the repeatability was 100%,The 536 sera of commodity swine in Xining Meat Processing Factory wereexamined with this method,the positive detective rate was 4.29%.The ex-periment proves that this method accords with the principles of conven-(?)ence,speediness,sensitivity,accurateness and thrifty.It can be easilyspread to the basis units.
