几种生物材料对人表皮细胞DNA合成的影响 被引量:3

摘要 目的 探讨Ⅰ型胶原、壳聚糖和海藻酸钠对人表皮细胞DNA合成的影响 ,为其在创伤创面的应用提供实验依据。方法 采用正交试验方法 ,以体外培养的人表皮细胞为实验模型 ,用3 HTdR掺入法分别测定各实验组3 HTdR掺入值 ,以反应其DNA合成水平的变化。结果 Ⅰ型胶原、壳聚糖、海藻酸钠处理组3 HTdR掺入值均较对照组明显升高 (P <0 .0 1) ,Ⅰ型胶原、壳聚糖与海藻酸钠合用可显著提高人表皮细胞3 HTdR掺入值 (P <0 .0 5 )。结论 Ⅰ型胶原、壳聚糖和海藻酸钠对人表皮细胞DNA合成具有促进作用 ,Ⅰ型胶原、壳聚糖与海藻酸钠合用对人表皮细胞DNA合成具有协同作用 ,提示Ⅰ型胶原、壳聚糖和海藻酸钠在皮肤创伤创面的治疗中具有应用前景。 Objective To study the effects of type Ⅰ collagen, chitosan and alginate on the DNA synthesis in human epidermal cells in vitro for providing experimental basis of their application in treating wound.Methods Cultured human epidermal cells were treated with type Ⅰ collagen, chitosan and alginate respectively according to the orthogonal trial approach. The changes of levels of DNA synthesis in human epidermal cells were determined by 3HTdR incorporation method. Results The 3HTdR values of human epidermal cells in type Ⅰ collagen, chitosan and alginate treated groups were significantly increased than that in control group(P<0.01). Combined collagen with chitosan and alginate could obviously increase the value of 3HTdR incorporation (P<0.05).Conclusion Type Ⅰ collagen,chitosan and alginate can promote the DNA synthesis in human epidermal cells.Combined collagen with chitosan and alginate can provide synergistic action in DNA synthesis in human epidennal cells.It shows that collagen, chitosan and alginate process the application prospect in treating skin wound.
