摘要
以溶液的表面张力为主要动力 ,辅以显微操作法去除一定量的卵母细胞胞质。测量了去除的细胞质体的直径 ,第一组为 38.6 4 μm ,第二组为 6 1.5 2 μm。小鼠卵母细胞的平均直径为 80 μm ,则第一组细胞质体占卵母细胞体积的 11.3% ,第二组细胞质体占卵母细胞体积的 4 5 .5 %。去除细胞质后两组卵母细胞均被氯化锶激活 ,其培养后卵裂率没有显著差异 (80 .0 %vs 77.9% ) ,两者都显著低于未经去质处理的激活卵母细胞和受精卵的卵裂率。两者的囊胚发育率虽没有显著差异 (37.3%vs 36 .8% ) ,但均显著低于未经去质处理的激活卵母细胞和受精卵的囊胚发育率 (4 8.3%和 73.3% )。去除 11.3%和 4 5 .5 %细胞质的卵母细胞孤雌激活胚、未经去质处理的激活卵母细胞和受精卵 ,体外培养的囊胚细胞数 (17.2 8± 7.16 ,11.5 0± 6 .83,2 1.86± 7.5 2 ,4 4 .6 7± 6 .86 ) ,各组间差异显著。
The effect of reduction of cytoplasm quantity on the in vitro development of parthenogenetic embryos from oocytes activated by Sr 2+ was studied in this experiment. Certain quantity of cytoplasm was squeezed out of the oocyte by using surface tension as main strength in micromanipulation. The diameters of cytoplasts and oocyte were measured. The average diameter was 38.64 μm in group 1, 61.52 μm in group 2, respectively. The diameter of oocyte was 80 μm. Thus the proportion of reduced cytoplasm quantity is about 11.3% in group 1, 45.5% in group 2 according to the calculation of global volume. The cleavage rates (80.0% vs 77.9%) of parthnogenetic embryos derived from activated oocytes reduced 11.3% and 45.5% cytoplasm were not different significantly, both of them were significantly lower than those of parthenogenetic embryos derived from activated normal oocytes or zygotes from in vivo. Similarly, the blastocyst rates(37.3% vs 36.8%)of these two parthnogenetic embryos from activated oocytes with reduced cytoplasm were not different significantly, both were significantly lower than those of parthenogenetic embryos derived from activated normal oocytes or zygotes from in vivo. The cell number in blastocyst of parthenogenetic embryos from activated oocytes reduced 11.3% and 45.5% cytoplasm, normal oocyte and 1-cell embryo from in vitro were different significantly between every two groups (17.28±7.16,11.5±6.83,21.86±7.52,44.67±6.86).
