高效液相色谱法测定锁阳中儿茶素的含量

Determination of catechin in Herbe Cynomorii by RP-HPLC

目的 建立锁阳中儿茶素的含量测定方法。方法 高效液相色谱法测定 ,ZORBAXRx -C18柱 (4.6mm× 2 5 0mm ) ,0 .0 4mol·L-1枸橼酸溶液 N ,N 二甲基甲酰胺 四氢呋喃 (5 5∶8∶2 )为流动相 ,流速 1.0mL·min-1,检测波长为 2 80nm。结果 测定了 2 0个不同产地和购置地的锁阳药材和饮片 ,药材的儿茶素含量 (以干品计 )为 2 .0 79%～ 0 .0 76 % ,饮片的含量仅为0 .30 0 %～ 0 .0 2 1%。平均回收率为 10 0 .2 4 %。结论 本法简便、准确、重现性好 ,可作为该药材和饮片的含测标准。

OBJECTIVE: To establish a RP-HPLC method for the determination of catechin in Herbe Cynomorii. METHODS: The ZORBAX Rx - C18 column was used. The mobile phase was consisted of 0.04 mol&middotL citric acid-N, N-dimethylformamide-tetrahydrofuran (55:8:2). The flow rate was 1.0 mL&middotmin-1 and the detection wavelength was 280 nm. RESULTS: Twenty samples were determined. The average recovery and RSD was 100.24% (n = 5) and 0.64%. CONCLUSION: The method appeared to be simple, accurate and reliable. It can be used as a quantitative method for quality control of Herbe Cynomorii.
