摘要
目的 从山茱萸中分离山茱萸多糖 ,并研究其理化性质及生物活性。方法 山茱萸经热水抽提、Sevag法脱蛋白、乙醇沉淀、DEAE 纤维素柱色谱分离 ,再通过SephadexG 2 0 0柱色谱进一步纯化 ,得到白色粉末状多糖PFCAⅢ。对该多糖进行完全酸水解、薄层色谱、红外光谱分析 ,并研究其对油脂抗氧化性能及清除自由基能力。结果 PFCAⅢ由鼠李糖、阿拉伯糖和葡萄糖以α 糖苷键组成 ,对油脂具有一定的抗氧化能力 ,并对Fenton体系产生的羟基自由基及邻苯三酚自氧化体系产生的超氧阴离子自由基具有一定的清除能力。结论 PFCAⅢ对油脂具有抗氧化性 ,并可有效清除羟基自由基和超氧阴离子自由基。
OBJECTIVE To study the isolation and physicochemical properties and bioactivity of polysaccharide PFCAⅢ from Fructus Corni .METHODS Polysaccharide PFCAⅢ was isolated from Fructus Corni through water extraction,deproteining,ethanol precipitating,DEAE-cellulose column chromatography respectively and was further purified by Sephadex G-200 column chromatography.Its component and antioxidation activities were analyzed.RESULTS PFCAⅢ was chiefly linked by α-glycosidic bonds.Chemical component analysis with TLC and HPLC showed that it was composed of rhamnose,arabinose and glucose in a molar ratio of 0.353∶1.299∶0.918.CONCLUSION PFCAⅢ showed antioxidation activity to axunge and gingili.It also has an action to scavenging hydroxyl free radical and superoxide anion free radical.
