摘要
建立了临床中毒患者血浆中甲基苯丙胺(MA)及其代谢产物苯丙胺(AP)的高效液相色谱 二极管阵列检测器(HPLC DAD)联用的定性定量方法。用固相萃取小柱分离、纯化、富集血浆中的MA和AP,在C8柱上测定,条件为以乙腈 甲醇 磷酸二氢钾缓冲液为流动相,流速为1 0mL/min,二极管阵列检测波长为210nm,检测范围为200~260nm。利用保留时间、紫外光谱进行定性分析。结果表明,血浆中MA和AP分别为0 036~1 438mg/L与0 047~1 500mg/L时线性关系良好;其回收率分别为96 1%~105 2%和92 1%~104 3%(n=5),日内相对标准偏差(RSD)与日间RSD均符合方法学要求;紫外最大吸收波长均为206nm。方法简单、快速。
A method for the simultaneous determination of methamphetamine and its metabolitesamphetamine in the patients' plasma of clinical poisoning by high performance liquid chromatography coupled with diode array detection has been established. Plasma samples were enriched, extracted and purified with solid phase extraction column and then analyzed on a CLCC8 column using a mobile phase of acetonitrilemethanolphosphoric buffer (25 mmol/L KH2PO4) at a flow rate of 10 mL/min. The detection wavelength was set at 210 nm and the detection range was 200-260 nm. Good linearity was obtained for the concentrations of methamphetamine and amphetamine in the range of 0036-1438 mg/L and 0047-1500 mg/L, respectively. The average recoveries were 961%-1052% and 921%-1043% (n=5), respectively. Their maximum wavelength was 206 nm. The method is suitable for the detection of methamphetamine and amphetamine in the plasma of the patients of clinical poisoning.
